A convenient and promising synthetic route to 4-amino-substituted 3-fluoroalkyl (aryl) -1,2-benzisoxazoles has been developed, which is subsequently used to obtain amines and lactams with various fluoroalkyl substituents. It has been found that radical Huang initiation conditions with Na2S2O4 can be used by adding a series of bromodifluoro-containing reagents and ethyl 2-bromo-2-fluoroacetate via vinyl esters. Obtained acetals can be considered as promising blocks for organo-fluorochemical chemistry which was demonstrated by the use of ethyl 4,4-diethoxy-2,2-difluorobutanoate for the multigram synthesis of 3,3-difluoro-GABA. The bromination reaction of α-methyl-substituted trifluoroacetylene with elemental bromine and NBS was investigated. The developed method allows to obtain new multifunctional α-bromomethy in multigram amount. The reactivity of this class of compounds with different nucleophiles and binucleophiles was investigated using the CF3-containing enone. The similarities and differences of the chemical properties between the newly obtained enones and the previously published analogues containing β-bromomethyl were shown. New promising synthetic methods for synthesis of polyfluoroalkyl-containing bioactive heterocycles were developed and new fluoro-containing heterocycles were synthesized and characterized: trifluoromethyl dihydroisoxazolinol and trifluoroacetyl 6H- [1,3] -thiazine.
