The optimum conditions for the lactate oxidase immobilization in the production of the lactate-sensitive biosensor were selected. The effect of solution parameters on the functioning of the biosensor (pH, buffer capacity and ionic strength) was investigated. The developed lactate biosensor showed good reproducibility of responses (standard error of determination did not exceed 1.4%) and high operative stability of signals throughout the week. The linear range of lactate determination was from 5 to 1000 μM, with a minimum lactate determination limit of 3 μM, and a sensitivity of 204 nA / mm. A methodological approach for the analysis of glutamate concentrations in samples of rat brain synaptosomes was developed. The presence of a basic signal in the preparation is shown in the absence of exogenous glutamate, which indicates the presence of endogenous glutamate in the samples. Baseline extracellular glutamate level was 7.9 ± 0.6 μM. The rate of glutamate accumulation (1 min) and the glutamate accumulation rate (10 min) were also determined by two biosensor methods and radioisotope analysis.
