The conditions for amplification of the introns of the α-, β-tubulin and actin genes in A. thaliana, O. sativa, L. and sitatissimum, S. tuberosum and S. lycopersicum using the appropriate species-specific primers were selected. The polymorphism of the intron lengths of α-, β-tubulin and actin genes in higher plant species was analyzed and specific DNA profiles were identified in different plant species and genotypes, which allowed them to be effectively differentiated. The same efficiency of using species-specific DNA markers to detect the polymorphism of the length of the introns of actin genes was shown in comparison with SSR markers by the example of analysis of genetic diversity in tomato varieties. It is established that the developed marker systems make it possible to quickly and reliably differentiate individual genotypes and plant species, and therefore they can be considered as a promising and effective tool for molecular genetic analysis.
