Kavetsky Institute of Experimental Pathology, Oncology and Radiobiology National Academy of Sciences of Ukraine popup.description2  In the framework of an experimental study using different incubation conditions in vitro, influencing the mitochondrial metabolism of tumor cells, some features of glucose metabolism in the Lewis lung carcinoma cells associated with their metastatic potential, namely the important role of reactive oxygen species in the resistance of metastatic active cells to anoikis. It was shown that LLC tumor cells with high metastatic potential are characterized by a high level of ROS production, which did not change in response to the deficit of oxidative phosphorylation substrates in their microenvironment. Unlike LLC, LLC/R9 tumor cells with a low metastatic potential generated high levels of ROS in response to the deficit of oxidative phosphorylation substrates in their microenvironment. The analysis of the electron-transport chain of mitochondria in tumor cells with various metastatic potential has been found to the progressive increase of nitrosyl hem and non-heme iron complexes during tumor growth in the case of low-metastatic LLC/R9. In contrast to the case of highly metastatic LLC, their level remained unchanged during tumor growth. The obtained results are the first stage in the elaboration of a scientific concept regarding the role of reactive oxygen species in the resistance of metastatically active malignant cells to anoikis. The scientific concept, which is elaborated, is aimed at developing new methods and drugs for inhibition of metastatic processes at an early stage of malignant tumor progression.
