This effect is explained by much faster accumulation of doxorubicin and cisplatin in the nuclei of the tumor cells compared to the native drugs due to the membranotropic action of fullerene C60, and leads to a much higher level of apoptosis induction in the tumor cells. Use of C60 fullerene-drug complexes also partially circumvents multiple drug resistance to doxorubicin and cisplatin in human HL-60 leukemia cells caused by overexpression of P-glycoprotein. Similar results were obtained in experiments in vivo. C60 fullerene-drug complexes resulted in a 2-fold increase in the therapeutic effect of doxorubicin and cisplatin towards murine Lewis lung carcinoma, decrease in the number and volume of tumor metastatic nodes, and a decrease in tumor-induced cachexia. This indicates that the intrinsic antioxidant activity of C60 fullerene not only reduces the potential side effects of drugs, caused by ROS overproduction, but also reduces the inflammatory processes caused by the growth of experimental tumors in mice and, as a consequence, improves the prognosis for animal survival.
