Objective: to determine the level of PCNA expression, MAPK kinases activity, amount of cleaved (89 kDa) PARP, and RET/PTC1/3 and BRAFWT/V600E mRNA expression in normal tissues, benign and malignant thyroid tumors; to study the level of p38 and Akt expression, the transcription factor c-fos, as well as the state of the hypothalamic-pituitary-adrenocortical system under the action of methanandamide and potassium ions in adrenal cortical cells. Research methods and equipment: protein electrophoresis, Western blot analysis, enzyme immunoassay, «Місro 22R» centrifuge, pipette dispensers, «SPU 123» electronic scales.Results: the expression level of PCNA, Akt, BRAFWT/V600E and siRNA-146b in tumor tissue of papillary thyroid carcinoma was higher, ERK1/2 – lower, and PARP did not differ compared to conditionally normal tissue. The intensity of RET/PTC1 or RET/PTC3 expression, as well as the presence of both rearrangements indicates tumor aggressiveness and invasiveness. Studies of RET/PTC1 and RET/PTC3 expression may be additional molecular markers of aggressive papillary thyroid carcinomas. Methanandamide was found to increase the expression of protein kinases p38 and Akt in the adrenocortical tissue of female rats in vitro. A single dose of 0.2 and 0.02 mg/kg methanandamide administered to intact rats resulted in increased activity of angiotensin-converted enxyme in the hypothalamus and adenohypophysis, and its decrease in the adrenal glands, and blood plasma. Corticosteroids in plasma were increased under their high doses. The multidirectional effects of total cholesterol content in the serum different-sex rats have been shown under single methanandamide dose. The changes detected may be related to the methanandamide ability to modulate the function of adrenocortical cells.
