The aim of the work: development of new bioanalytical methods for the quantitative determination of creatinine – a metabolite of protein metabolism and important biomarker of functional state of the kidneys using as an analytical tool recombinant form of stable creatinine deiminase (CDI) from microbial source. The aim of the 2019 stage: Screening, physicochemical and structural characterization of polymeric and nanocomposite materials suitable for the immobilization and stabilization of creatinine deiminase and chemosensing the ammonium ions. Research methods: microbiological, biochemical, physicochemical, electrochemical, enzymatic. The screening of nanocomposite electron-transfer mediators for the chemosensing the ammonium ions was performed and Cu/Zn(Hg)S nanoparticles, as well as, electrodeposited polyaniline (PANi) in complex with Nafion have been selected. The physicochemical and structural characterizations of the nanocomposites have been performed. The conditions of immobilization of CDI onto chemocomposite-modified surface of different types of working electrodes have been optimized. On the basis of Cu/Zn(Hg)S nanoparticles and immobilized CDI, a laboratory prototype of amperometric creatinine-selective biosensor has been constructed. The biosensor is characterized by high selectivity, sensitivity (90.8 ± 2.8 А∙М-1∙м-2), and a wide linear range (0.03 - 3.5 mM) to creatinine. On the base of PANi/Nafion membrane with immobilized CDI a highly sensitive (423.5 ± 20 А∙М-1∙м-2) creatinine biosensor with a lower limit of determination up to 12 μM and a linear range 0.015 – 0.95 mM of creatinine has been constructed. According to the results of the project for 2019, were published 10 articles (with a total impact factor of 10.93), 2 abstracts of conference reports and submitted 1 article.
