The high-performance SPR spectrometer is designed to simultaneously perform many biological analyzes, which is an ideal tool for probing large micro-arrays of biomacromolecules and for investigating their interactions with various partner molecules. The second stage of this project was to investigate the efficiency of the process of immobilization of thiolated oligonucleotides on the sensor surface of bioselective elements of hybridization DNA sensors in a medium of citrate buffer solution (pH 3) of different ionic strength and compare with the results of traditional method in KH2PO4 (pH 4)). In the process of developing approaches to modify the sensor surface of the spectrometer by biomacromolecules, the effect of pH and ionic strength of the medium on the level of immobilization of thiolated oligonucleotides on the gold surface was investigated. It has been shown that the level of immobilization of mod-Ph thiolated oligonucleotides on the sensor surface is directly proportional to the concentration of immobilization solutions. It was found that the process of hybridization of P1 targets with mod-Ph immobilized probes elicits sensor responses whose magnitude demonstrates their dependence on P1 concentration (in the range up to 200 nM) close to linear for all types of bioselective elements under study. The use of a medium with a lower pH (pH 3) did not significantly increase either the level of immobilization of thiolated oligonucleotides or the level of hybridization of the target with the immobilized probes.   Product Description popup.authors popup.nrat_date 2020-07-03 Close
