Karazin Kharkiv National University popup.description2  According to the technical assignment of the fourth stage of the project, interaction of fullerenes and bacteriophage MS2 in solutions with electrolytes and with organic dyes, as well as encapsulation of europium coordination complexes into the liposomes were investigated. Specifically, novel monomethine cyanine probes Cl-YO, F-YO, Cl-YO-Et, Cl-YO-Bu and YO-Pent were investigated for their ability to detect and characterize MS2 using fluorescence techniques. The tested compounds showed significant fluorescence response to the presence of the intact bacteriophage at physiological values of ionic strength. Furthermore, association of the cyanines with the MS2 surface was suggested, as evidenced by a decrease in the fluorescence intensity of the fluorophores in the presence of MS2 under the influence of denaturants, urea and guanidine hydrochloride. Significant stabilizing effect of electrostatic interactions in the probe-MS2 complex at low ionic strength values (10 mM) was also revealed. The above spectral properties of the novel cyanine compounds allowed to consider them as promising agents for studying changes in the structure of the virus under the influence of heat (Cl-YO-Et, Cl-YO-Bu), denaturants (Cl-YO, F-YO), and ionic forces (for all investigated compounds). Next, using pyrene and diphenylhexatriene, we demonstrated that encapsulation of the europium coordination complexes in liposomes is accompanied by changes in the membrane free volume and in the packing density of the lipid molecules. The results of this research will be useful in organic synthesis, in pharmacology, e.g. for the development of the effective therapeutic strategies against cancer and against diseases caused by “superbugs”.
