Aim of the study: to investigate the effects of natural alkaloids homoharringtonine, licorine, and berberine on the proliferation, migration and invasion of tumor cells of different tissue origins, as well as the molecular mechanisms of the realization of their antitumor effects. Research methods: proliferative potential was assessed using MTT test and colony formation test, migration - using in vitro scratch test, invasion - using a modified Boyden chamber. Changes in gene expression levels were investigated by quantitative PCR and Western blot analysis. It was found that the use of homoharringtonine led to decreased levels of proliferative activity of MDA-MB-231breast cancer cells , increased expression of caspases-3, -8 and -9, N-cadherin, VEGFR2, GSK3β, β-catenin, WNT signaling inhibitors DKK1, 2 , 4. Licorine was found to inhibit proliferation and migration of breast cancer cells lines MDA-MB-231 and MCF-7, and to modulate the expression of E- and N-cadherins. Berberine reduced the proliferative potential, motility, and invasiveness of mouse LLC lung cancer cells, along with the decreased expression of adaptor protein Ruk/CIN85 and the cancer stem cells marker CD44.
