Objective: to investigate the relative level of gene expression of the ATM/CHEK2/P53/PUMA cascade and associated genes-candidates in the peripheral blood leukocytes of Chornobyl clean-up workers depending on age, doses of exposure, quantitative genomic instability parameters and the presence of oncological pathology. Methods of research: immunological, molecular genetic, cultural, statistical. Results. Manifestations of genome instability were found in Chornobyl clean-up workers and persons with oncological pathology by the relative telomere length, the distribution of lymphocytes over the phases of the cell cycle, the expression of histone gamma-H2AX and the Cyclin D1 protein. Relationships between the quantitative radiation-induced genomic instability parameters with the expression of genes associated with the ATM/CHEK2/P53/PUMA cascade, which are involved in the regulation of inflammatory processes (TNF, IFNG), telomere length (TERF1, TERF2, TERT), apoptosis ( BCL2, HNRNPA1), antioxidant protection (GSTT1) and oncological transformation (TP53, EGFR, VEGF) in Chornobyl clean-up workers. The markers of genome instability in the remote period after irradiation and early diagnosis of oncological transformation were determined.
