The subject of the study is to determine the parameters of binding of some drugs to human serum albumin by quenching luminescence. Research methods - spectrophotometry, fluorescence spectroscopy. The main idea of the work was to expand the capabilities of the luminescent method of analysis to determine the binding of drugs (D) to human serum albumin (HSA). It is shown that D quench the intrinsic fluorescence of the protein as a result of static interaction in the HSA-D system. It is established that the quenching of luminescence is carried out due to the non-radiative transfer of electron excitation energy (FRET) from the luminescent substance (donor) to the quencher (acceptor). Binding parameters are defined: constants, number of binding sites and distances between donor (HSA) and acceptor (D). Based on the calculated thermodynamic parameters of D binding to HSA, it was found that Van der Waals interactions and the formation of hydrogen bonds play an important role. The study of conformational changes of HSA caused by binding to D by measuring synchronous fluorescence spectra. It is established that for almost all D there are changes in the conformation of the protein near the tryptophan residue. A highly sensitive method for the determination of propoxazepam in rinse from the surfaces of pharmaceutical equipment has been developed. It is shown that the intrinsic fluorescence of some drugs is manifested in the same region of the spectrum as the intrinsic fluorescence of the protein, and therefore its emission spectrum is superimposed on the intrinsic fluorescence of the protein. In this case, the possibility of determining the binding constants of HAS with drug molecules by quenching their own fluorescence was demonstrated for the first time.   Product Description popup.authors Yegorova Alla V.
