Purpose: development of new bioanalytical methods for quantitative determination of creatinine (Crn) - a protein metabolite, an important biomarker of the kidneys functioning using as an analytical tool recombinant form of stable creatinine deiminase (CDI) from microbial source. The purpose of the 2020 stage: construction of a laboratory prototype of a portable biosensor device for the analysis of Crn, compatible with modern gadgets. The main tasks of the stage included: 1) construction of ammonium-selective chemosensory membrane based on electricallytical conductive polyaniline (PANi); 2) developing a bioelectrode based on recombinant CDI and ammonium-selective nanocomposite PANi film; 3) construction of a microcurrent potentiostat integrated with a bioelectrode and a chemoselective membrane, suitable for producing of Crn analyser. Research methods: microbiological, biochemical, physical, physicochemical, physico-mathematical, electrochemical, enzymatic. As a result, the design and testing of a miniaturized microcurrent potentiostat compatible with modern gadgets was developed. The device is adapted for use with commercial planar electrodes modified by recombinant CDI. Software for controlling a miniaturized microcurrent potentiostat in combination with modern gadgets based on the Windows operating system has been created. The suitability of using the constructed developed producer portable potentiostat for various types of amperometric measurements (cyclic voltammetry and chronoamperometry) is demonstrated.
