Section 3. Development of reference methods for the arginine analysis with the usage of arginine-hydrolyzing recombinant enzymes. popup.stage_title Head Sybirnyi Andrii A., Registration Date 07-01-2021 Organization Institute of Cell Biology popup.description2  Report about scientific investigation: 41 pages, 13 figures, 3 tables, 61 references. Objects of study: enzymes of bioanalytic importance – recombinant bacterial arginine deiminase, ammonium-sensitive nanomaterials with electrono-conducting activity. The purpose of scientific investigation (section No3, ICB of NAS of Ukraine) is to develop reference methods of arginine analysis using arginino-hydrolyzing recombinant enzymes. The purpose of the 3rd stage of the work (section No 3) in 2020 is the development and characterization of electrochemical biosensors based on arginine deiminase and nanomaterials. Research methods: microbiological, biochemical, physicochemical, analytical. As a result of the work carried out in 2020: A number of nanomaterials possessing electrono-conductivity were synthesized, investigated and screened on their sensitivity to ammonium ions. On the base of the most effective nanomatherials the amperometric chemosensors on ammonium ions were constructed and their analytical characteristics were studied. Amperometric biosensor for arginine analysis was developed using recombinant arginine deiminase (ADI) and the best ammonium-sensitive chemosensor. The proposed ADI-biosensor was tested on the real samples of juices in comparison with referent enzymatic-chemical method (with the usage of arginase I), that was developed by us earlier. High correlation was demonstrated for results of both biosensor and reference method. 2 papers and 2 abstracts were published, 1 paper and chapter in monograph were submitted. The work was done according to the plan.
