Study Purpose − evaluation of the effects of trophic, hormonal, osmogenic and gelling components of nutrient media, determination of the efficiency of the improved technology for androgenic haploid induction, production of spring barley haploids and populations of doubled haploid lines for validation of smut disease resistance genes. Study Methods – anther culture in vitro, culture of isolated embryos, light microscopy, methods of variation statistics and analysis of variances (ANOVA). Theoretical and practical results – For the first time, a comparative study of mannitol for spike cold pretreatment and as a supplement to inductive medium was carried out. The both methodical approaches were shown to be applicable for spring barley haploid production in anther culture in vitro. For the first time, an effective chemically modified starch-based medium for plant regeneration was elaborated. The expediency of combination of the improved mode for spike pretreatment at a low above-zero temperature (patent Nj 113261, 26.12.2016) and cultivation of anthers on nutrient media containing new formulas of chemically modified starches instead of agar was proved. 1,497 androgenic green plants and their seed progenies – populations of doubled haploid lines for validation of smut disease resistance genes were produced from F1−F3 hybrids of 17 cross combinations. A patent for the invention “The Mode of Spring Barley Immature Embryo Culture in Vitro Production” (N o 118385, 10.01.2019) was received.
