Bogatsky National Academy of Sciences of Ukraine popup.description2 The objects of study – Serratiopeptidase Serratia E15, polyvinyl alcohol, chitosan, sodium alginate, sodium salt of carboxymethylcellulose. The purpose of the work is to develop gels and gauze wipes with immobilized serratiopeptidase, evaluation of immobilization results by the level of complex proteolytic activity and study of biochemical and physicochemical properties of the obtained drugs. The proteolytic enzyme Serratia sp. E15 with high caseinolytic, collagenolytic and fibrinolytic activities; the homogeneity of the enzyme (MM 45 ± 4 kDa) was proved by SDS-electrophoresis. New biocompatible highly active polymeric materials of complex proteolytic action of long-term storage in the form of gauze bandages with serratiopeptidase included in polyvinyl alcohol /sodium alginate and gel based on sodium salt of carboxymethylcellulose have been developed. The physicochemical characteristics of the obtained products were studied; shown the absence of significant differences in pH profiles, the expansion of the thermoprofile in the region of elevated temperatures in the case of dressings compared to the free enzyme. Immobilized preparations were found to be 5.1 and 2.9 times (for dressings and gels, respectively) more thermostable compared to free enzyme. It was determined that the immobilization of serratiopeptidase does not significantly affect Vmax. hydrolysis. An increase in KM of 1.2 and 1.3 times, as well as an increase in the inhibition constant Ki of 3.5 and 4.7 times, respectively, is shown. Product Description popup.authors Dekina Svitlana S.
