It was found that the contractile response of a cell varies depending on the degree of its mechanical stretching, and the action of oxytocin and TRPC4 ion channel agonists differs from that without mechanical stress. These data are significant in the context of the use of ion channel agonists as pharmacological means of normalizing the function of the myometrium. The study developed an algorithm for quantitative analysis of uterine contractions, which consists in the use of templates for automatic search of peaks Amplitude, halfwidth, frequency, area under the curve and time constants of rise and decay are suggested to be determined, as well as the contribution of these parameters to the work produced by the muscle. It was found that the calix[4]aren C-1087, the inhibitor of Ca2+,Mg2+-ATPase activity of myometrium cells plasma membrane, stimulates the increase of Ca2+ concentration in the smooth muscle cells cytoplasm, which does not depend on the state of pregnancy. These data are the basis for further studies of calix[4]arens as selective inhibitors of Ca2+,Mg2+-ATPase activity of myometrium cells plasma membrane, as well as for the development of pharmacological uterotonic agents on their basis for correcting the contractile function of the uterus at pathological conditions. The ATP concentration-dependent increase of ionized calcium concentration in mitochondria matrix in the absence of exogenous Са2+ was registered. Several evaluations were made, thus the Hill coefficient equals 3,18 ± 0,27 and activation constant for ATP equals 0,97 ± 0,07 mM.
