Designs of promising gRNAs for activation of human cardiogenesis genes by CRISPR-VPR system have been developed. All sequences were synthesized and cloned into a plasmid for Cas9sgRNA expression (Addgene # 68463). A plasmid expressing fusion proteins of catalytically inactivated Cas9 and transcription activation domains - VPR was also developed. Therefore, a collection of plasmids expressing selected gRNA sequences was created for further molecular genetic analysis and in vitro validation. In general, the data obtained by us are a necessary and logical basis for studying the prospects of using the CRISPR system for cell reprogramming for therapeutic purposes.
