The purpose of scientific work is to create a biochemical screening test system to determine the state of the blood-brain barrier to determine the quantitative markers of post-traumatic syndrome of various genesis, correlation justification for changes in the amount of neurospecific proteins in serum and cerebrospinal fluid depending on changes in BBB permeability. Applied methods: experimental models of intracranial hemorrhage and focal ischemia, physiological tests of animal behavior, biochemical methods for the study of specific proteins and enzymes, organotypic culture of the hippocampus, statistical methods of data analysis. The distribution of adhesive and cytoskeletal proteins specific for BBB and brain (glial fibrillar acidic protein (GFAP), calcium-binding protein S100B, neuronal molecules of cell adhesion, etc.) in the brain and serum of experimental animals has been established. A correlation analysis of changes in animal behavior and changes in the content of neurospecific proteins in the brain and serum was performed. It was found that the violation of mnestic processes and the development of oxidative stress in experimental animals with reproduced in the experiment acute cerebrovascular pathology are associated with changes in the content of various forms of neurospecific proteins in the brain. For the first time an analysis was performed between the content of water-soluble forms of the studied proteins and their membrane or filamentous forms for different types of post-traumatic condition. It is proposed to use for the study of BBB not only experimental models in vivo in laboratory animals, but organotypic brain cultures in vitro also.
