The object of research is MCF7 breast cancer tumor cells, 2D and 3D cell models in vitro. The goal is to adapt the method of cultivation of multicellular tumor spheroids to the maintenance of primary cultures from tumor tissues of patients and to obtain mammospheres in vitro, to determine the effect of the conditions of the cellular microenvironment on the population of tumor stem cells in mammospheres. Research methods - cyto-morphological, microscopic, flow cytometry. As a result of the research, methods were developed for enriching the population of tumor stem cells of the standardized MCF-7 breast cancer line in monolayer and spheroid culture. Methods for enriching a subpopulation of cells with stem characteristics and analysis of the resulting cells were identified and tested. The results of the study indicate an increase in the expression of stem cell markers such as CD24, CD44, CD133, Bmi1 protein, EpCam in enriched multicellular tumor spheroids as compared to monolayer growth and conventional BPS. Scientific and practical value.
