Karazin Kharkiv National University popup.description2 A detailed theoretical and experimental study of the interaction of the antimicrobial peptide gramicidin S with model membranes of different morphology (flat multilayer and bilayer spherical) and lipid composition was performed, the influence of GS on the structural and functional characteristics of blood cells and connective tissue was carried out. The mechanisms to control the membrane activity of GS are revealed. Two types of GS binding to lipid membranes corresponding to the binding of GS monomers and oligomers have been identified and characterized. Thermodynamic parameters of binding were calculated. A sharply increased affinity of GS for the cholesterol-enriched phase was found in membranes with a low cholesterol content (10 wt.%). Dehydration of phosphate groups of phospholipids during binding to GS has been reported. The degrees of hydration and redistribution of water molecules between free and bound states in gramicidin GS complexes with liposomes of different lipid composition were determined. The degrees of hydration of cryoprotectants for the development of ways to reduce the hemolytic effect of GS on erythrocytes were calculated. The presence of D-g-PAA (PE) polymer modulates the interaction of GS with the DPFH membrane, reducing the overall membraneotropic effect and promoting the binding of GS oligomers to the membrane in expense of monomers. Computer simulation of lipid membranes of different composition and their complexes with GS on bilayer membrane models was performed. Gramicidin S at concentrations from 5 to 20 μg / ml reduces the ξ-potential of intact erythrocytes, primarily by partially neutralizing the negative charge of sialic acids by positively charged amino groups of ornithine residues.
