The aim is to elucidate the mechanisms and create a physical-mathematical theory of cells cryo-damage by electrical voltages that occur in the cell suspension during its low-temperature preservation, and to develop effective ways to prevent these damages. Research methods – physical-mathematical analysis and numerical simulation (computers), light microscopy, cryomicroscopy, morphometric analysis (cryomicroscope, confocal microscope). The physical-mathematical model of substances transmembrane transfer between cells and the environment is built, taking into account the electrical membrane potential. The computer program has been created for calculating the dynamics of changes in cell volume, cryoprotectant concentrations, basic ions and transmembrane potential during the cell exposure in cryoprotectant solution and freezing to eutectic temperatures. An algorithm for determining the filtration and permeability coefficients for cryoprotectants of cell membranes at subzero temperatures has been developed. The model of electrical breakdown of cell membranes is created, taking into account short-acting hydration repulsive forces between lipid bilayers. The physical-mathematical model based on the two-factor theory of cryodamage, thermodynamic theory of homogeneous crystallization and general theory of activation-type processes has been developed for determining the probability of cell damage by solution effects and intracellular crystallization to finding the optimal cooling rate.
