Synthetic optimization of the structure of hit-compounds was performed on the basis of predicted and experimental screening data. The optimal conditions for the passing of alkylation reactions of thiosulfonates with bromoalkyl phthalimide derivatives are developed and the influence of the reaction medium on the yield of the target products was investigated. It was found that the best yields of S-(2-(1,3-dioxyisoindolin-2-yl)alkyl esters of 4-acetylamino-, 4-amino-, 4-methylbenzenethiosulfonic acids were obtained in acetone medium at Prolonged boiling of the reaction mass, new derivatives of benzenethiosulfonic acid salts with chromene fragments were synthesized and identified. A one-pot approach to the synthesis of new dithiocarbamate derivatives of 2-chloro-N-(9,10-dioxo-9,10-dihydroanthracenyl)acetamide was proposed. The relationships of the influence of temperature, the medium and the position of the 2-chloro-N-acetamide substituent in the 9,10-anthracenedione ring on the formation of the products of thiocyanation of 2-chloroacetamides of 9,10-dioxoanthracene were established.
