The aim of the study was investigation of adamantane derivatives antibiofilm activity general mechanisms. Complex of modern physico-chemical, biochemical, microbiological, histochemical, microscopic, molecular, pharmacological and statistical methods was used in the study. It was established for the first time that compounds with adamantyl (AM-166, KVM-97) and N-alkylaryl (KVM-194) radical disrupted the biofilms formation of S. aureus, E. coli, and P. aeruginosa, inhibited the expression of ica and icaA genes and caused genomic variability in S. aureus (revealed 3 DNA fragments in the range of 400-500 bp and variability in the range of 1000-2000 bp). The influence of compounds on biofilms was associated with changes in the components of the matrix, the ability to influence persister cells. The adamantane-containing compounds disrupted QS-dependent processes in P. aeruginosa (synthesis of pyocyanin, hemolysins, and autoinducers), and blocked efflux pumps in E. coli which was confirmed by experiments with ethidium bromide and genes tetA and tetR expression in E. coli.
