It was found that the use of lysate of tumor cell (LDC) line A549, enriched in PSC, significantly enhances the level of co-expression of CD86 and HLA-DR molecules on the generated dendritic cells (DC) compared with DCs loaded with LDC native line A549, p < 0.05. The load of the culture of DC LPC line A549, enriched in PSC, contributed to a significant increase in the number of CD83 + cells compared with the culture of DC without antigen loading, p <0.05. It was proved that the loading of DCs enriched with PSK line A549, contributed to their Tx1-polarization, namely statistically significantly increased expression of mRNAIL-12 and IFN-γ, and led to a significant increase in the expression of chemokine CCR7, indicating an increase in the migratory activity of DC and the ability to initiate an immune response (p <0.05). According to the results of research, the load of DCs of the A549 line enriched with PSC does not induce suppressor functions in the generated DCs - the expression level of IDO, IL-10 and TGF-β mRNA remains at the level of DCs loaded with the A549 line. The obtained results substantiate the possibility and prospects of using the components of cell lines enriched in PSC to optimize methods for obtaining mature DCs suitable for use as a cell base in the creation of individual therapeutic antitumor vaccines with increased antitumor efficacy.
