During the reporting period, a screening of the effect of a combination of classic anticancer drugs and newest compounds on the biochemical and cytological parameters of monolayer and spheroid cultures was carried out. Object of research: multicellular tumor spheroids (МTS) of the MCF-7 line, monolayer culture of the MCF-7 line, salinomycin, doxorubicin, methotrexate, cisplatin. The work involved cytological, molecular biological methods, morphometric and statistical methods of analysis. The aim of the study was to study the effect of anticancer drugs, which are often used in the clinic and affect actively proliferating tumor cells in combination with the potential anticancer drug salinomycin in different types of tumor cell cultivation (adhesive and suspension). Determination of proliferative, morphological, kinetic characteristics of spheroids in monolayer culture under incubation conditions with a combination of anticancer drugs. The objectives of the study were to investigate differences in the sensitivity of tumor cells after incubation at different concentrations and different combinations of anticancer drugs. Determine the most effective combination of drugs for monolayer and suspension culture of MCF-7 cells. It was found that anticancer drugs (doxorubicin, methotrexate, cisplatin) in combination with salinomycin demonstrate a noticeable cytotoxic and antitumor effect. With a combination of drugs with different mechanisms of action, it was demonstrated that the synergistic effect of anticancer drugs with salinomycin is more noticeable than the individual action of drugs. At the same time, the population of tumor stem cells turned out to be sensitive to this effect.
