Purpose of the first stage in 2021 year – synthesis of reactive monomeric dyes-intercalators in quantities sufficient for further preparation of dimeric dyes. Obtaining of reactive probes suitable for the conjugation with the oligonucleotides. Preparation of dimeric dyes-intercalators. Comparison of properties of monomeric and dimeric dyes-intercalators under PCR conditions. Research objects – monomeric and dimeric dyes-intercalators based on derivatives of acridine orange and monomethine cyanine dyes; FLT-sensitive fluorescent dyes of different classes. Research methods – organic synthesis, spectral and other physical and photophysical research methods – NMR-spectroscopy, mass-spectrometry, electron absorption and fluorescence spectroscopy. Results obtained at the first stage of the project in 2021 year: – Methods for the synthesis of reactive monomeric dyes-intercalators based on the derivatives of acridine orange and asymmetric cyanine dyes were improved. The synthesis of monomeric dyes in quantities sufficient for further production of dimeric dyes-intercalators was carried out. – Developed and improved methods for obtaining structural analogues of commercial dyes-intercalators SYBR Green I and YOYO-1. – The synthesis and purification of reactive FLT-sensitive dyes of various classes of fluorescent compounds were carried out. – The structures of the synthesized FLT-sensitive dyes were established, and their spectral and photophysical properties were studied. – Methods for the synthesis of dimeric dyes-intercalators have been developed. – Data on the spectral and photophysical properties of the obtained dimeric dyes in solutions were obtained.
