Based on the industrial lots of «DIA®-Brucella-IgG Enzyme immunoassay for the detection of anti-Brucella IgG", a significantly improved kit of reagents for the serological diagnosis of brucellosis in humans was obtained by replacing the conjugate containing the G Streptococcus protein with a more active and specific conjugate containing monoclonal antibodies to human IgG labelled with HRP. The most effective conjugate Mab 153H11-HRP as well optimal pairs of specimen and conjugate diluents were selected. During the research the ELISA procedure was significantly optimized by developing calibrators that allow the semi-quantitative and quantitative analysis of human serum/plasma samples for IgG to the causative agent of brucellosis, which significantly increases the accuracy and quality of the results. «DIA®-Brucella-IgG" and analogues of leading manufacturers were compared using brucellosis positive and negative sera, the shortcomings of the industrial kit "DIA-Brucella-IgG" were identified as well the approaches to overcome them in a new test kit - a candidate for a prototype.
