Aim: To investigate molecular mechanisms of action of amphiphilic drugs on lipid membranes of viral particles and cells infected with viruses, their mechanisms of sensitivity to membraneotropic drugs. Evaluation of the possibility of selective action on viral membranes using membrane-tropic drugs and the design of new drugs. Methodologies: The model objects will be: strongly curved artificial membranes liposomes, artificial lipid bilayers, membrane membranes of nerve cells, artificial liposomes. The research will be carried out by IR and Raman spectroscopy, confocal microscopy, fluorescent membrane probes, computer modeling by molecular dynamics. Novelty: For the first time explored the influence of membraneotropic drugs on physical characteristics, parameters of structural organizations of lipid membranes, similar in composition, physical properties to viral membranes and plasma membrane of human cells, its prospects for antiviral therapy, the ability to achieve selectivity of membraneotropic action on the membrane. During the second stage: The influence of cholesterol and hyper- / hypothermia on the interaction of drugs with the lipid membrane of nerve terminals and model bilayers was determined. Possibilities of selective influence on viral membranes by means of membraneotropic drugs on the basis of physical differences of viral and cell membranes are estimated. Recommendations for the design of new drugs that will have a greater impact on viral membranes have been developed. The influence of membraneotropic drugs on pore formation in a flat bilayer membrane was determined. The effect of drugs on key characteristics of synaptic transmission in the nerve terminals of the brain of rats: accumulation and release of glutamate and GABA, exocytosis, membrane potential, acidification of synaptic vesicles, spontaneous and stimulated generation of reactive oxygen species.
