The research deals with actinobacterial genes for pleiotropic regulators of AraC and RpoZ families, kinases and pathway-specific regulators of StrR, SARP, LuxR families. Research goal is to explore mechanisms of transcriptional regulation of cryptic genes for secondary metabolism; to apply these genes for discovery of novel bioactive compounds using the menioned above Streptomycetales and Micromonosporales species. A network of transcriptional regulation genes for secondary metabolism has been described for the studied species , as well as the diversity of gene clusters for antibiotic production. Genome of landomycin A producer S. cyanogenus S136 has been sequenced. S. albus J1074 strain deficient in adpA gene has been isolated. S. roseochromogenes strain overproducing clorobiocin have been developed. A library of plasmids for expression of regulatory genes has been developed. For certain recombinant strains there was revealed a production of novel bioactive compounds, whose structure has been elucidated. Evidence was provided to support the structural assignments for these novel structures, some of which belong to derivatives of polyene lucensomycin and aminocoumarin clorobiocin.
