The purpose: to determine the role of mitochondrial redox balance and non-specific mitochondrial pore activation (mPTP) in the development of diabetic cardiomyopathy and the possibility of its correction with natural antioxidants. It has been established that metabolic disorders such as insulin resistance, hyperglycaemia and dyslipidemia cause mitochondrial dysfunction in cardiomyocytes by reduction of succinate dehydrogenase activity and inhibition of aconitase activity. It has been determined that mitochondrial dysfunction in cardiomyocytes is accompanied by a disturbance of the redox balance in the heart mitochondria. The development of mitochondrial dysfunction and oxidative stress in mitochondria induces an increase in the sensitivity of cardiomyocytes to proapoptotic factors. It has been determined that mitochondrial dysfunction in cardiomyocytes disrupts the functional state of the cardiovascular system, in particular, changes the heart rhythm and causes the development of systolic and diastolic dysfunction. Quercetin was found to inhibit the development of mitochondrial dysfunction in cardiomyocytes by increasing metabolic activity and oxidative capacity of mitochondria. Quercetin prevents the development of oxidative stress by reducing the production of reactive oxygen species and increasing the activity of antioxidant enzymes in the cardiomyocytes of diabetic rats. Quercetin increases the resistance of myocardial cells to proapoptotic factors. The inhibitory effect of quercetin on the development of mitochondrial dysfunction in cardiomyocytes determines its cardioprotective properties, confirmed by the normalization of the heart rate and suppressing of the diastolic myocardial dysfunction manifestations in diabetic rats.
