Polymers BP83-1 and F8DM1 also efficiently delivered the plasmid DNA pFlag-P21WT, which encodes gene of the p21 protein, to HEK293T, HeLa, U251 and MCF-7 cells. Optimal concentrations of polymers for delivery of plasmid DNA to cells have been identified. Polymeric carriers BP83-1 and F8DM1 showed significantly higher efficiency of delivery of the P53wt gene in HEK293T and HeLa cells, compared with such efficiency of polyethyleneimine. Transfection of cells with polymexes of polyDMAEMA carriers BP83-1 and F8DM1 with pEGFP-N1-p53 or pFlag-P21WT led to a significant inhibition of cell viability of HeLa cells, which can be explained by changes in cells due to the expression of delivered genes of p53 and p21. An increase in the number of HeLa cells in the G2 phase after their transfection with the BP83-1 / pFlag-P21WT complex was detected, which indicates the blocking of these cells in the G2 / M phase of the cell cycle. The obtained results indicate a high potential of polyDMAEMA as carriers for the delivery of genes of interest to mammalian cells.
