The aim of this study is to identify genes associated with metastasis in patients with malignant brain tumors and to identify factors that affect their regulation. Research methods - isolation of genomic DNA, RNA, siRNA and proteins from brain tumors and adjacent tissues, bisulfite conversion, methyl-specific PCR, quantitative real-time PCR, Western blot analysis, statistical analysis. As a result of the project, the change of molecular genetic markers of oncogenesis in human glioma tumors was assessed: expression and production of TSC22D1, ERVW-1, RUNX3, CHI3L1 genes and probable influence on their regulation by methylation of promoter regions and deactivation of PN microfluids. We determined that the production of CHI3L1 protein is the most characteristic feature of gliablastoma of III and IV degree of malignancy. The correlation between the level of mRNA and RUNX3 gene protein in tumors with changes in the methylation of its promoter, as well as the increase in methylation of the promoter of the GRX3 gene in tumors is shown. They found elevated levels of miR-21-5p, miR-24-3p, miR-30a-5p siRNA in glioblastomas compared to control samples, which makes them potential biomarkers of brain tumor progression.
