At the same time, down-regulation of Ruk/CIN85 led to the opposite consequences. The modulating role of Ruk/CIN85 in the control of expression of reprogramming genes and genes of markers of cancer stem cells was also established. Simultaneously, overexpression of Ruk / CIN85 in 4T1 and MCF-7 cells has been shown to significantly increase the expression/activity of key enzymes/components of glycolysis and, conversely, to reduce the activity/expression of the enzymes of oxidative phosphorylation. Moreover, in 4T1 cells with up-regulation of Ruk/CIN85 a significant increase in the levels of activity/expression of enzymes involved in extracellular matrix remodeling, control of invasive potential and chemoresistance, enzymes/molecules involved in the control of redox status were also demonstrated. For most parameters, reverse changes were observed in 4T1 cells with knockdown of Ruk/CIN85. The identified patterns of changes in the transcriptome and metabolome of breast adenocarcinoma cells for the first time showed the involvement of adaptor protein Ruk/CIN85, depending on its expression levels, in the control of dedifferentiation/differentiation processes, and EMP associated with epithelial-mesenchymal-amoeboid/amoeboid-mesenchymal- epithelial transitions.  Product Description popup.authors Gerashchenko Olga Leonydivna Horak Iryna Iryna R.
