The object of research is the process of obtaining a highly active virus-safe preparation of coagulation factor VIII using affinity chromatography on macroporous silica sorbents. The aim of the work is to develop a method for obtaining a highly active virus-safe preparation of coagulation factor VIII with the use of biospecific silica sorbents with immobilized active dyes and study of its properties. Research methods: protein fractionation; gel and ultradiafiltration; spectrophotometric; electrophoretic; affinity and ion exchange chromatography; coagulological; antivirus processing; enzyme-linked immunosorbent assays. Main scientific and practical results: the scheme of obtaining the FVIII / vWF complex from blood plasma was created, which includes the use of solvent-detergent and thiocyanate antiviral treatment, cryoprecipitation, salt and PEG fractionation, ion exchange chromatography on DEAE-Sepharose and and negative affinity sorption on macroporous silica sorbents; the maximum degree of purification of FVIII from 239 to 702 times, depending on the type of sorbent; obtained a preparation of factor VIII with a specific activity of 90.28 ± 1.74 IU/mg protein, the ratio of FVIII/vWF:Rcof 1.73 ± 0.21; no residues of non-target proteins were detected in the preparation; the stability of the preparation under different storage regimes was studied. Field of application: blood centers, chemical and pharmaceutical companies, research organizations. Efficiency. The implementation of research results will improve the technology of obtaining factor VIII coagulation preparations from donor plasma by increasing efficiency, reducing time, minimizing losses of factor activity in the purification process.
