For this purpose several variants of insertional cassette were constructed and introduced into the genome of O. polymorpha recipient strain. The selection of transformants with increased colony size was performed on the minimal medium with 15% L-arabinose. The ability to produce ethanol from xylose and L-arabinose was evaluated in selected transformants. As a result strains with increased ethanol production from xylose and L-arabinose were obtained. Using standard molecular techniques the insertional cassette along with flanked regions was isolated from genomes of selected strains with increased ethanol production. The identification of disrupted genes involved in fermentation of xylose in O. polymorpha will be performed by sequence analysis of isolated DNA fragments.
