Kavetsky Institute of Experimental Pathology, Oncology and Radiobiology National Academy of Sciences of Ukraine popup.description2 Intensification of glycolysis has been shown for the first time to be one of the mechanisms that ensure the survival of metastatic cells during the transition to deadhesive growth and form their resistance to anoikis. The degree of resistance depends on the ratio of the activity of the antioxidant system and the functioning of the mitochondrial system. Defects of the mitochondrial system correlate with a more pronounced sensitivity of cells to anoikis and low metastatic potential. Inhibition of glycolysis by oxamate (LDH inhibitor) in vitro causes a concentration-dependent cytotoxic effect on LLC and LLC/R9 cells, more pronounced for LLC/R9 cells. Administration of this inhibitor to mice with LLC was not accompanied by inhibition of growth and metastasis of this tumor due to stimulation of tumor angiogenesis resulting from the concentration-dependent effect of oxamate on the functional polarization of macrophages.
