Study Purpose – improvement of bioassays for laboratory evaluating gibberellic activity of new agents with supposed gametecidal capacity via selection of the most appropriate test-objects, determination of optimal testing regimes, and comparison of results of laboratory investigations with those of sunflower inflorescence treatment in the field. Study Methods – laboratory techniques and field methods. Barley and corn seeds were used for production of test objects. Bioassays based on growth response (leaf elongation in cut coleoptiles) of prepared corn seedlings as well as on qualitatively determination of α-amylase activity induced in aleurone layer of endosperm in response to exogenic gibberellin treatment were improved. The optimal corn seedling length for production of test object was determined. 0.005 % aqueous solution of gibberellic acid was used for the treatment of corn seedlings, embryo-free barley seeds and sunflower inflorescences in the field. For the first time, it was proposed to use hulless barley varieties for production of test object (embryo-free seed halves) for determination of gibberellic activity. Absence of hull allows for simplification of the embryo dissection procedure. For the first time, the advantage of using seeds from barley varieties with waxy endosperm was proved. In particularly, it was revealed that the bioassay time was reduced from 6-7 to 4 days due to higher bioavailability of starch with a low amylose content for hydrolysis by α-amylase released under the influence of exogenous gibberellin. It was shown that further acceleration (up to 2 days) in the laboratory assessment could be achieved by immersion of embryo-free seeds in gametocidal agent solutions for 24 h with a subsequent one-day storage in Petri dishes containing moist filter paper.
