Registration Date 24-01-2022 Organization Institute of Molecular Biology and Genetics of NAS of Ukraine popup.description2  The purpose of the second stage of the project (the second stage in 2021) is is to develop an in vitro model of the catheter-associated infection caused by K.pneumoniae and to study the effects of azithromycin and colistin on this model. The following tasks were set in accordance with the goal. The first task was to construct an in vitro model of the catheter-associated infection caused by K.pneumoniae. The second task was to apply AZM-combination therapy for the treatment of K.pneumoniae infection in an in vitro model. The third task was to investigate the structure of K.pneumoniae biofilm under the influence of AZM-combination therapy. In the third stage of the project, an in vitro model of a catheter-associated infection based on a collagen matrix seeded with the HEK293 cell line and inoculated with UHI1667 hospital isolate was developed. The developed in vitro model was used to study the effect of AZM-combined therapy. The structure of K.pneumoniae biofilm under the influence of AZM-combination therapy in the developed model was studied, in particular, inhibition of amidoid fibril production in infectious K.pneumoniae UHI1667 biofilms in the presence of AZM was observed.
