The research aim was to determine the ways of implementation of antitumor action of hybrid nanocomplexes against Ehrlich carcinoma cells by their ability to generate the reactive oxygen species and modulation of the expression rate of tumor cell stemness genes. Modern research methods were used in the work: flow cytometry, real-time polymerase chain reaction, light microscopy. It was found that the antitumor effect of hybrid nanocomplexes was a multistage process, accompanied by mitochondrial dysfunction and as a consequence it stipulated the generation of reactive oxygen species and subsequent induction of apoptotic / necrotic changes in tumor cells. Simultaneously with such changes there was a decrease in the expression rate of stemness genes (nanog, oсt4, sox2) and the corresponding transcription factors (Nanog and Oct 3/4). That is, the synthesized nanocomplexes are compounds that simultaneously cause cell death by both apoptosis and necrosis. It is noteworthy that, the decrease in the expression rate of the studied stemness genes after treatment with nanocomplexes of the heterogeneous population of EAC occurred against the background of a reduction in CD44+ - cells with enhanced tumorigenic potential, which emphasizes the unique role of this receptor in a direct activation of stemness genes.
