The patients with PMF were divided regarding to the DIPSS system into risk groups: "intermediate-2" (13 patients), "intermediate-1" (5 patients), low risk (7 patients). Molecular genetic studies were performed in 22 patients with PMF, 16 of which were JAK2-positive. Mutations of CALR and MPL genes were analysed in JAK2-negative patients, and a mutation in CALR (CALR -del52bp in two and CALR -ins5bp in one case) was revealed in 3 patients. Three patients were triple negative. Significant fluctuations of the VEGF, TNF-alpha, IL-6, IL-8, IL-1beta and IL-10 levels were revealed in patients with MF, PV and ET. In the group of patients with MF, the level of VEGF was significantly higher compared to the control group. In the analysis of VEGF levels in patients with PV, the median VEGF was more than 3-fold higher than in the control group. Analysis of statistical and clinical parameters revealed a close correlation between VEGF levels and platelet counts. No relationship has been established between VEGF and other laboratory parameters, including the presence of the JAK-2 mutation. VEGF levels in patients with ET were higher than in the control group. The median VEGF level in patients with ET was more than 4 –fold higher than in healthy people. Analysis of the levels of proinflammatory cytokines (TNF, IL-6, IL-8, IL-1beta) and anti-inflammatory cytokine (IL-10) also found a significant difference between each of the subgroups in MF and ET for all 5 analyzed cytokines, whereas in PV patients a significant increase in concentration was revealed only for interleukin-6 (p = 0.012).
