Information × Registration Number 0222U001510, 0121U112551 , R & D reports Title Myogenic mechanisms of urinary bladder mechanical sensitivity in norm and type 2 diabetes. popup.stage_title Head Shuba Yaroslav М., Доктор біологічних наук Registration Date 28-01-2022 Organization Bogomolets Institute of Physiology of the National Academy of Sciences of Ukraine popup.description2  It is established that: 1) both urothelial cells and detrusor smooth muscle (DSM) cells of the bladder of rats express protein of all three types of mechanoactivated cation channels - PIEZO1, PIEZO2 and TRPV4; 2) induction of type 2 diabetes does not result in obvious changes in protein expression of these channels; 3) isolated DSM cells by type of membrane current, which is activated in response to mechanostimulation (hydrodynamic shock), could be subdivided into three groups: 57% of the cells did not generate any current, 27% generated non-selective cationic current with inward and outward components and 16% generated outward, potassium-selective current; 4) the chemical activator of PIEZO1 channels, Yoda1, caused an unusual response in DSM cells in the form of partial blockade the outward current component without any signs of activation of the inward current; 5) the use of chemical activator of TRPV4-channel, GSK1016790A, was ineffective in terms of activation of any current; 6) the outward mechanoactivated potassium current in DSM cells is transported through TREK-1 channels, as this current could be blocked by L-methionine and activated by arachidonic acid. The data obtained are contradictory. On the one hand, the protein of PIEZO1, PIEZO2 and TRPV4 channels is present in urothelial and smooth muscle cells, and, on the other, the DSM cells do not respond to chemical activators of these channels.
