Materials and methods. Cryopreservation, lyophilization, flow cytometry, spectrophotometry, polymerase chain reaction. Results of the research. Cryopreservation of human cord blood leukoconcentrate was accompanied by redistribution of cell composition towards an increase in the number of hematopoietic stem cells (CD34+CD38-, CD34+CD38+). Lyophilization led to an increase in the content of T regulatory (Treg) cells and the average and total fluorescence intensity of CD25 marker, which might indicate an enchancement of immune-suppressive activity compared to native and cryopreserved biomaterial. In animals with adjuvant arthritis (AA), the benefits of concomitant use of lHCBL and cPE compared with monotherapy in the treatment of joint injury and immune disorders have been shown to improve indices of pro- and anti-inflammatory cytokines (TNFα, IL-6, IL-17A, IFNγ, IL-10), structural and functional indices of Treg cells. The combined use of lHCBL and cPE increased the amount of Treg cells and the total fluorescence intensity of CD25 markers in animals with AA, which could be considered as a promising way to strengthen the anti-inflammatory mechanism and restore autotolerance in autoimmune pathology.
