The aim of the study was to investigate the gonadoprotective efficacy of the combined use of metformin and metabolic drugs (lipoflavone and metovitan) in the metabolic syndrome that developed during the juvenile period of development of male rats. Research methods - toxicological, pharmacological, biochemical, molecular biological, morphological, statistical. The use of metformin in the induction of MS in male juvenile rats in the distant period of development did not have a positive effect on the indicators of the structural and functional state of the gonads. Co-administration of metformin with metabolic drugs, lipoflavone or metovitan, helped to reduce disorders of differentiation and maturation of germ cells, the restoration of sperm count. Increased expression of CYP3A2 mRNA involved in testosterone metabolism in the testes of rats with MS and the absence of a positive effect of metformin indicate impaired testosterone metabolism. Administration of metformin with metabolic drugs decreased CYP3A2 gene expression and normalized serum testosterone levels. CYP2E1 gene expression was also reduced, testicular antioxidant status was restored, and embryolethality was reduced. It has been experimentally proven that the use of metformin in MS from juvenile age is not able to effectively prevent disorders of the male reproductive system, which requires its joint use with drugs of metabolic action.
