The aim of the study was to identify molecular weight distribution of peptides in rats’ normal serum and under myocardial ischemia and necrosis in order to determine the role of endogenous peptides in regulating inflammation and remodulation of the heart after injury, and to establish the mechanisms of cardioprotective action of tissue-specific heart peptides. Materials and methods. Electrocardiography (ECG), spectrophotometric, spectrofluorimetric, gel permeation chromatography, matrix-assisted laser desorption/ionization (MALDI), histological. Research results. Molecular weight spectra of the peptides in serum depend on the age of the animals. The obtained data confirm the possibility of identifying native serum and serum without thermolabile proteins, and monitor changes in their composition using synchronous fluorescence spectra. The decrease in the fluorescence intensity of the probes after ischemia and myocardial necrosis indicates an increase in the competition of peptides for binding sites on albumin molecule. On day 28 there is a significant increase in fluorescence intensity of the probes, indicating a decrease in the binding sites occupied by the peptides. It has been shown that fluorescence intensity of K-35 probe depends on the time elapsed after myocardial ischemia, and this approach can be used to diagnose myocardial necrosis and stage of the process. Using MALDI method, it has been found that after myocardial necrosis certain peptides, which are absent in the norm, are registered in the serum. The biological activity of the extract is manifested in the fact that when it is administered to animals with cryonecrosis of myocardium, the diameter of necrosis decreases compared to animals that have not received treatment.
