It was shown for the first time that the combination of a low-carbohydrate diet and metformin can effectively inhibit the growth of solid tumors in mice, including highly metastatic melanoma B16. This effect was accompanied by zero mortality of tumor-bearing mice even on the 100th day after the start of treatment, and complete normalization of the functions of hematopoietic and immune systems in experimental animals. However, for the complete elimination of the tumor, a minimum dose of the traditional anticancer drug should be additionally used. It was found that the glycolysis inhibitors 3-bromopyruvate and sodium hydroxycitrate do not show a therapeutic effect towards murine NK/Ly lymphoma model, and the combination of 3-bromopyruvate with a low-carbohydrate diet generally showed an antagonistic effect. It was also established that the modulation of energy metabolism against the background of long-term starvation for arginine does not lead to an additive effect in inhibiting the growth of tumor cells. The combination of a low-carbohydrate diet, metformin, and glibenclamide together with low doses of anticancer drugs (in particular, doxorubicin) showed a pronounced synergistic effect and led to a 4-fold extension of life in tumor-bearing animals with NK/Ly lymphoma and L1210 leukemia. The obtained results indicate significant prospects for the further application of the proposed scheme of treatment of cancer patients in clinical practice.
