Protocols for adipogenic (white, bright/beige, brown fat) differentiation of human MSC-LC using nutrient media that do not contain blood serum (serum-free media) were developed; and nutrient media with a completely chemically defined composition (chemically defined media).2. It was noted that there were no significant morphological changes in the MSC-VT between the groups with the classic and modified environment. It was established that the chemical composition of both inductive media provides support for the vital activity and metabolic activity of MMSC-LT during adipogenesis at a consistently high level, and the level of extracted dye (Red Oil O), gene expression profiles, and cytokine production are equally high in both comparison groups. 3. The dynamics of changes in the morphofunctional properties of human MSCs during cultivation in nutrient media with a changed D/H isotopic ratio, as well as the peculiarities and efficiency of adipogenic differentiation at different deuterium content in the nutrient medium, were studied. A change in almost all indicators (morphological changes, proliferative capacity, cytotoxicity, migration activity and metabolic activity) was established under conditions of increased (>150 ppm) deuterium content in the growth medium. At a deuterium content below the natural value (150 ppm), a stimulating effect on proliferative activity was observed, but deuterium depletion (<150 ppm) caused cytotoxicity. 4. In vitro test systems have been developed based on human MSCs with and without serum / fully chemically defined media using different ratios of deuterium/protium in the culture medium. The possibility of using these test systems for the screening of pharmacological drugs based on deuterium for the treatment of T2DM diabetes is shown.5.
