The purpose of the work is to analyze the sensitivity and selectivity of optical biosensor platforms and mechanisms of interaction with K. pneumoniae antigens. Authors identified and serotyped K. pneumoniae strains isolated from patients and carriers, and selected the most common antigenic types of microorganisms. The sensitivity of the isolated microorganisms to eleven antibiotics was determined and a high level of multiresistance among K. pneumonia strains was established. A powerful antibacterial and antibiofilm potential of silver nanoparticles against multiresistant K. pneumoniae was revealed. The theoretical and methodological bases of bioselective layers based on ZnO composites creation was improved. A method of the bioselective layer formation on the surface of ZnO nanostructures within the limits of covalent binding of specific antibodies has been developed. The efficiency of the deposition of the components of the bioselective layer was evaluated using specific immunological reactions. An experimental setup was created to study the optical properties of composites based on ZnO by the method of photoluminescence spectroscopy, and a developed sample chamber was used to study materials containing K.pneumoniae antigens. The intensity of photoluminescence was measured and the output signal of the measuring transducer was interpreted, which confirmed the possibility of determining K.pneumoniae in the studied sample in an amount exceeding 103 CFU/ml.
