Registration Date 20-12-2022 Organization Institute of Cell Biology of the National Academy of Sciences of Ukraine popup.description2  It was found that the covalent attachment of fatty acid residues to the 9th carbon of the berberine molecule enhances the cytotoxic effect of this alkaloid on tumor cells, and this effect is better expressed when a fatty acid residue is attached with a longer carbon chain length and a greater number of unsaturated bonds. The complexation of berberine and its covalently modified derivatives, as well as sanguinarine, which is structurally related to berberine, with a synthetic polymer (poly(PEG-co-GMA) (5:95) with a grafted PEG monomethyl ether fragment (Mn=750 Da)) and with a co-polymer of PEG with methacrylate and dimethyl maleate (poly(PEGMA)-co-DMM) (PEGMA-DMM)) was done. Such complexation made it possible to obtain water-soluble forms of berberine, its modified derivative, and sanguinarine. They possessed the antineoplastic effect on human colon carcinoma cells of HCT116 wt and HCT116 p53-/-. Therefore, the double modification of berberine carried out in this work improves its physicochemical properties and significantly enhances its antitumor activity.
