Purpose: to define the effectiveness and therapeutic activity mechanisms of bone marrow mesenchymal stromal cells in skin radiation lesions in rats on allogeneic and xenogeneic models. Methods: radiobiological, hematological, cell culture, morphological, statistical. Equipment: CLINAC 6 MeV linear accelerator, UNIDOS universal dosimeter, MN 120 lamina shafts, EC-160 CO2 incubator, microscope Leica 6000B LED, chambers Leіca DFC 310 FX, microscope Leіca DM ІL LED, microscope Axіolab 94D 2601, biochemical automatic analyzer Respons 910, ultralow-temperature refrigerator, analytical scales. Theoretical and practical results: the anti-inflammatory action of cultured/cryopreserved BM MSCs mechanisms have been established by the analysis more than 2.0-fold decrease the level of C-reactive protein in blood under the local and/or two-time systemic therapeutic administration during the skin inflammatory reactions after irradiation. The characteristics of the functional activity of cultured/cryopreserved BM MSCs were determined in terms of 80% viability, 8.5% colony formation efficiency, and adipogenic and osteogenic differentiation. The regenerative effect of locally administered 1.5 x 106 and 1 x 106 cultured/cryopreserved BM MSCs on days 14 and 21was determined by reducing the severity of skin clinical reactions, accelerating the skin wound healing and reducing the skin damage area by 1.2 times without radiation ulcers on day 42. Novelty: for the first time on the developed model of radiation skin lesions of varying severity in animals (rats) the anti-inflammatory and regenerative mechanisms and assessment of the therapeutic action of cultured/cryopreserved bone marrow mesenchymal stromal cells were evaluated. Use area: radiobiology.
